Cell free protein synthesis (CFPS) has become a powerful tool for protein expression in vivo. Escherichia coli is one of the most commonly used cell types in extract preparation, and it is also the first system developed for cell-free protein synthesis. After several improvements, E. coli cell-free expression system has become a powerful, cost-effective, multifunctional and scalable technology, which can be used to produce vaccines, antibodies, cytokines and other proteins, paving the way for the emerging field of cell-free metabolic engineering.

Creative BioMart optimized a simple and compact blue light induced E. coli startup system based on El222 protein and provide protein synthesis services based on the system.

How dose it Works?

El222 is a blue light controllable DNA binding protein. It belongs to the lov (light oxygen voltage) domain protein family. When exposed to light, it will change its form and bind to DNA.

Cell free reaction mixture: RNA polymerase holoenzyme complex, ribosome, aminoacyl tRNA synthase, translation factor and energy mixture with blue light induced DNA and pre translation el222 (blue light switchable DNA binding protein).

Our simple blue light inducible plasmid system based on El222. In short, when exposed to blue light, El222 dimerizes and binds to DNA. This may initiate transcription by recruiting RNA polymerase to the promoter region. El222 is inactive in the dark and therefore cannot bind DNA.

E. coli Extract Protein Synthesis Service

The basic principle of E. coli cell-free protein synthesis system

  • Plasmid DNA or PCR products are used to synthesize mRNA under the action of RNA polymerase.
  • The translation factors, enzymes, amino acids, energy and tRNA of E. coli extract were used to translate mRNA into protein.
  • mRNA is reused to synthesize protein in E. coli extract.
