Organelle Manipulation

The intracellular transport and distribution of organelles play an important role in a variety of cell functions, including cell polarization, intracellular signal transduction, cell survival and apoptosis. Through the dimerization of CRY2-CIB1, a common optical switch, the precise spatio-temporal control of light on the transportation and distribution of organelles can be realized. Technical Principle

CRY2 sequences were anchored to organelles through specific organelle targeting transmembrane domains by editing techniques.

Scheme 1: CIB1 is associated with truncated kinesin. Under blue light irradiation, cib1-cry2 binds to recruit the kinesin motor to the organelle, which drives the organelle to move to the positive end of the microtubule.

Scheme 2: CIB1 domain is linked to dynein / dynein adaptor protein. When irradiated with blue light, cib1 binds to CRY2 to recruit dynein into organelles. Therefore, organelles are pushed to the negative end of microtubules.
