With the progress of metabolic engineering and synthetic and systems biology, biological production has become an increasingly attractive and competitive strategy for the synthesis of biomolecules, recombinant proteins and biofuels from renewable raw materials. However, due to low productivity, it is still difficult to make biological production processes economically feasible on a large scale. By balancing the two characteristic stages of biological production, namely growth and production, this is the core of the trade-off that has a significant impact on productivity. The development of photogenetics provides the possibility for the optimization of biological production.

Creative BioMart has optimized / developed a variety of optogenetic systems for biosynthesis and protein expression. The versatility and controllability provided by these systems will be the key factor to achieve high-level protein expression in E. coli.

Optogenetic Applied Biological Compound Production

We provide biological production services for microorganisms, including but not limited to metabolites such as muconic acid, mevalonate acid or isobutanol, important enzymes such as β-glucosidase, or antimicrobial drugs.

Muconic acid expression solution

Blue light sensitive protein El222 was used to regulate the expression of CRISPRi system, which closed the competitive pathway and redirected the metabolic flux to heterologous muconic acid synthesis in E. coli.

Mevalonate expression solution

The system is based on the Lac operon system. We placed lacI under the promoter of pDawn plasmid, so that the genes of interest were inhibited under light and induced in the dark. The circuit aims to induce gene expression in the dark to avoid the potential challenge of limited light penetration in high cell density fermentation.

β-glucosidase expression solution

We will combine the SRRz autolysis system and light induction system of autophagy, which provides a new idea for the efficient and economic production of protein.

Antimicrobial drugs release platform

The synthetic drug is allowed to diffuse and deliver to the surrounding medium in a light dose adjusted amount. Using the focused beam, the synthesis and release position of drugs can be determined freely.

The main purpose of constructing these expression systems is to maximize the yield by adjusting the close balance between culture growth stage and biological production stage.
