Optogenetic is a technology involving the use of light to manipulate cell activities with high temporal and spatial accuracy in vitro or in vivo. The activity of selected cells can be turned on and off on a biologically relevant time scale of a few milliseconds. The field of optogenetics integrates optics and genetic engineering to measure and manipulate cells (usually neurons) and corresponding molecular processes. Compared with drugs and lesions, photogenetics has much higher specificity and control over single cells.

The main pillars of photogenetics experiments include biological components and photogenetics instruments. At present, we have four technical platforms, Neuroscience Technology Platform, Cell Strain Development Platform, Model Organism Development Platform and Optogenetic Equipment Platform.

Construction of Biological Components

The key first step of photogenetics experiment is to select the appropriate photogenetics probe.

Here are some key factors to consider

  • Photogenic excitation or photogenic inhibition
  • The color of the active light
  • Stimulation time
  • Opsin delivery system

These questions will help you select the appropriate photogenetic probe for the experiment.

Once you choose a photogenetic probe, it must be expressed at the target site, usually through virus expression and transgenic animal models.

The research objects of photogenetics are diverse, which can be divided into organisms (model organisms) and cells. Cells can be divided into nerve cells and non nerve cells.
