Technology Principle

Optogenetic-T7 E.coli system –Creative BioMart

We photoinduced T7 RNA polymerase by dividing the polymerase into two fragments and fusing them into a light-activatable dimerization domain. After light induction, the two domains dimerize, so that the N-end of one domain is spatially close to the C-end of the other binding domain. This allows these domains to be fused to the C-terminal of the N-terminal fragment of the mitotic protein and the N-terminal of the C-terminal mitotic fragment, and the enzyme is spatially reconstructed by incorporating photogenetic regulation into the T7 RNA polymerase and assembled and dissociated by light stimulation.
