C. Elegans Knock-in Services

Caenorhabditis elegans is widely used in genetic and biomedical research. DNA fragment insertion can be introduced into the genome of Caenorhabditis elegans. Creative BioMart's Caenorhabditis elegans knock in services include point mutations, floxed alleles, degron markers, fluorescent markers and immune markers. In the past few years, we have produced several fluorescent transgenic lines, including different fluorophores and GFP variants.

  • C. elegans Point Mutation Service

Our point mutation service uses CRISPR / cas9, which is the best gene editing method to create small and accurate editing to introduce small nucleotide changes at the target site.

  • C. elegans Fluorescent Tagging Service

C. Elegans are particularly suitable for fluorescence studies because they are transparent. Fluorescence can be easily observed in both real-time and fixed images.

  • C. elegans Immunotag Service

Immunolabeling was added using CRISPR / cas9 gene editing technology. All tags were optimized for expression in Caenorhabditis elegans. Optional labels include FLAG、HA、HIS、TAP or S-peptide.

You can choose from a variety of degron inducible and developmental systems, including auxin inducible degradation, photosensitive or blue light inducible degradation, degron labeled membrane topology reporter gene and oma-1 label, which are used to degrade proteins in 1 cell stage.

We carefully designed our humanization project to retain endogenous transcriptional signals, so as to maintain expression patterns and levels. In addition, we have a proprietary sequence optimization protocol that we use to create a transcript that can be well expressed in Caenorhabditis elegans.

Step Description
Transgenic design Selection of guide RNAs and/or design of donor homology plasmid or DNA oligonucleotide with special attention to codon usage, intron placement, and splicing.
Injections Injection of mix into the C. elegans gonad.
Candidate screening Initial screening. Identification of animals displaying selectable marker or co-CRISPR phenotypes.
Strain confirmation Confirmation of transgenic line(s). Screening of homozygous F2 animals via PCR and sequencing.
