The transgenic mice of this strain can be used for cardiovascular optogenetic research. A mutant calcium-translocating channelrhodopsin was inserted and expressed in the cardiomyocytes of transgenic mice. The expression of the mutant opsin can enhance the permeability of muscle cells to Ca2+, accelerate the reaction kinetics and more than 50 times the light sensitivity. Therefore, irradiation of cells expressing mutant channelrhodopsin with blue to green light (~ 450-530 nm; maximum 474 nm) will lead to rapid and reversible stimulation of channel opening (for example, calcium inflow and contraction of muscle, or strong action potential discharge activity in neurons). Because it is possible to activate the protein under naturally occurring light intensity while maintaining high time accuracy, it may also be used in gene therapy and visual restoration.

Strain Description

After the cardiomyocytes of the transgenic mice were irradiated by blue to green light, the mutant calcium transport channelrhodopsin molecule will lead to the reversible stimulation of channel opening, calcium inflow and muscle contraction.

Bacterial opsin is a retinal binding protein that binds photosensitive domains and ion channels or pumps. It provides light dependent ion transport, membrane potential change and sensory function for bacteria. Chlamydomonas reinhardtii derived channelrhodopsin-2 (ChR2 or COP4) is a blue light driven cation channel, which can depolarize cells and generate action potential.
