The transgenic mice of this strain can be used for optogenetic research, and provide a useful tool for the induced light stimulation of glutamatergic neurons involved in rhythm generation and motor initiation. The channel rhodopsin-2 cDNA sequence of Chlamydomonas reinhardtii was modified to include codons optimized for mammalian expression and function acquisition sequence replacement. The optimized channelrhodopsin-2 sequence was fused with enhanced yellow fluorescent protein (EYFP) sequence. Transgenic mice expressed channelrhodopsin-2/EYFP fusion protein targeting the spinal cord and hindbrain motor region.
Under blue light, ChR2 activates YFP expressing cells and induces neuronal activity of glutamatergic populations in the spinal cord and hindbrain motor regions.
