Strain for Sensorineural Research

The transgenic mice of this strain can be used in optogenetic research to quickly control motor behavior by adding or removing light.

The channel rhodopsin-2 cDNA sequence of Chlamydomonas reinhardtii was modified to include codons optimized for mammalian expression and function acquisition sequence replacement in order to produce greater fixed photocurrent. The optimized channel rhodopsin-2 sequence was fused with the yellow fluorescent protein sequence and inserted downstream of the mouse tryptophan hydroxylase 2 (Tph2) promoter.

Opsin is a retinal binding protein that provides light dependent ion transport and sensory functions for halophilic bacteria. The optimized opsin acts as a blue light driven cation channel to depolarize cells and generate action potential. Therefore, irradiation of expressed neurons with blue light (450-490 nm) will lead to rapid and reversible stimulation of action potential discharge/neural activity in these cells. Hemizygous mice have the ability of survival and fertility, and the life expectancy is normal. In vitro and in vivo studies of neural circuits/connectivity after illumination, and fluorescent labeling of serotonergic neuronal populations.

After opsin expression is activated by light, this strain of mice can be used for in vitro and in vivo study of neural circuit/connectivity after illumination, or for fluorescent labeling of serotonergic neurons.

Expressed Gene: Yellow Fluorescent Protein (YFP) from jellyfish, Channelrhodopsin from Chlamydomonas.

Site of Expression:

The dorsal raphe nucleus, giant cell reticular nucleus, paramedian reticular nucleus and other serotonergic nuclei showed moderate YFP expression.
