Chlamydomonas reinhardtii derived channelrhodopsin-2 (ChR2 or COP4) is a blue light driven cation channel that depolarizes cells and generates action potential. The calcium-translocating channelrhodopsin (CatCh) is a ChR2 variant.
Transgenic mice of this strain expresses calcium-translocating channelrhodopsin CatCh in smooth muscle cells (such as blood vessels, lungs and airways). Bacterial opsin is a retinal binding protein that binds photosensitive domain and ion channel or pump, providing bacteria with light dependent ion transport, membrane potential change and sensory function. We optimized CatCh. The optimized CatCh shows stronger Ca2+ permeability, resulting in faster response time and voltage response to light than wild-type ChR2 Sensitivity is about 70 times.
Irradiation of CatCh molecules with blue to green light in smooth muscle resulted in reversible light stimulation of channel opening, calcium influx and muscle contraction.
CatCh has high sensitivity to blue to green light. Therefore, irradiation of CatCh expressing cells with blue to green light (450-530 nm, maximum 474 nm) will lead to rapid and reversible stimulation of channel opening (e.g., calcium inflow and contraction of muscle, or strong action potential discharge activity in neurons, etc.). Because CatCh activation is possible under naturally occurring light intensity while maintaining high time accuracy, it may also be applied to visual restoration in gene therapy.
