The environmental light required for plant growth often leads to unwanted system activation, which leads to the lag of photogenetics in plant research.
Arabidopsis thaliana is a popular model organism in plant biology and genetics. It is the first plant to sequence its genome. It is also a popular tool for molecular biology to understand many plant traits, including flower development and light sensing.
Creative BioMart has developed a photogenetic system based on Arabidopsis to control gene expression in plants inactive under white light and can be activated by monochromatic red light. The system includes two engineered photoreceptors, which exert combined activity in the regulation of transcription initiation: one inhibits gene expression under blue light and is modified by EL222 photoreceptor, and the other activates gene expression with red light and is based on phytochrome B (PhyB)–PIF6 optical switch.
This is an integrated optogenetic molecular tool, which consists of two parts: one module activates gene expression under red light and the second module ensures effective transcriptional inhibition under blue light. The basic principle behind this method is that the combination of two switches will produce a system that is inactive under ambient growth conditions (light and dark) and only active when irradiated with red light. This makes plants grown under standard light conditions fully applicable.
