Workflow for Generating Optogenetic C. elegans Strains

Step	Description
Transgenic design	Find the optimal promoter, coding and terminator sequences to maximize the chance of successful strain generation.
Molecular assembly	De novo sequence generation or gene assembly method to obtain the target sequence, and effectively generate the sequence components required to create efficient injection mixing components.
C. Elegans injections and Candidate screening	Perform a sufficient number of injections to ensure more than 3 Biological repetitions. Screening candidates.
Strain confirmation	Each transgenic junction was viewed by PCR to deliver a sequence validated strain.
