Strain Construction

The transgenic mice of this strain can be used for optogenetic research to study olfactory transduction, olfactory circuit and olfactory perception of light stimulation by adding or removing light.

Strain Description

The channelrhodopsin-2 cDNA sequence of Chlamydomonas reinhardtii was modified to include codons optimized for mammalian expression and function acquisition sequence replacement in order to produce greater fixed photocurrent. The optimized channel rhodopsin-2 sequence was fused with the enhanced yellow fluorescent protein sequence and inserted downstream of the mouse olfactory receptor promoter 160 (olfr160).

Bacterial opsin is a retinal binding protein that provides light dependent ion transport and sensory functions for halophilic bacteria. The optimized opsin acts as a blue light driven cation channel to depolarize cells and generate action potential. Therefore, irradiation of expressed neurons with blue light (450-490 nm) will lead to rapid and reversible stimulation of action potential discharge/neural activity in these cells. Hemizygous mice have viability and fertility, and their life expectancy is normal.

At present, Creative BioMart has developed a variety of mouse strains expressing Channelrhodopsin, halorhodopsin and Archaerhodopsin respectively, which facilities the application of optogenetic. We have complete animal maintenance facilities to ensure animal health and animal welfare.

Expressed Gene: Yellow Fluorescent Protein (YFP) from jellyfish, Channelrhodopsin from Chlamydomonas.

Expressed Site: The fusion protein was only expressed in olfactory neurons expressing specific odor receptor M72.
