The modified Archaerhodopsin-3 (Arch) from halorubrum sodomense was fused with enhanced green fluorescent protein (EGFP) and inserted into the endogenous locus of transgenic mice. The strain is Cre recombinase dependent. When mating with mice expressing Cre recombinase, the resulting offspring will delete the stop box in CRE expressing tissues; Results in the expression of Arch EGFP fusion protein. The strains of hemizygous mice were dynamic, fertile, normal in size, and did not show any obvious physical or behavioral abnormalities.
Irradiation of these cells with yellow green light (~ 575 nm) resulted in reversible inhibition of action potential discharge/neural activity in these cells. These transgenic mice can be used for optogenetic reseArch to express an inhibitory opsin, which can effectively inhibit the activity of cortical neurons (and other excitable cell types, such as muscle cells and immune cells). Compared with the improved Arch, the optical sensitivity is increased by more than 3 times, which is equivalent to an increase of more than 200% in the volume of brain tissue treated by a typical single optical fiber.

Compared with wild-type Arch, the improved Arch can mediate photocurrent with similar maximum amplitude, but compared with Arch, the photosensitivity is more than three times higher, which is equivalent to more than twice the volume of brain tissue treated by a typical single optical fiber. The expression of modified Arch on neuronal membrane and axon is good, which may support improved neural pathway silencing.
