Strains Expressing Halorhodopsin

The transgenic mice of this strain can be used for optogenetic research to express inhibitory opsin that effectively silences neuronal activity. The fragment formed by the fusion of halorhodopsin from natronomonas and enhanced yellow fluorescent protein (EYFP) was inserted downstream of the mouse thymocyte antigen 1 (Thy1) promoter of transgenic mice. The strains of hemizygous mice were dynamic, fertile, normal in size, and did not show any obvious physical or behavioral abnormalities.

Irradiation of halorhodopsin expressing cells with yellow to red light (~ 589-680 nm) resulted in reversible inhibition of action potential discharge/neural activity in these cells. These transgenic mice can be used for optogenetic research to express inhibitory opsin that effectively silences neuronal activity.

Strain Description

Bacterial opsin is a retinal binding protein that binds photosensitive domains and ion channels or pumps. It provides light dependent ion transport, membrane potential change and sensory function for bacteria. The mice of this strain used halorhodopsin as a yellow light driven chloride ion pump, which can lead to hyperpolarization and prevent action potential. Therefore, irradiation of halorhodopsin expressing neurons leads to rapid and reversible light inhibition of action potential discharge/neural activity in these cells.

These mice allowed in vivo light inhibition of cortical activity and limb movement. Halorhodopsin EYFP positive cells include neurons in various regions of amygdala, midbrain and lower brain stem, pyramidal cells in layer 5 of cortex, cells in anterior ventral nucleus of thalamus, granular cells in dentate gyrus and pyramidal cells in CA1 area of hippocampus, and mossy fibers in granular cell layer of cerebellum.

Irradiation of CatCh molecules with blue to green light in smooth muscle resulted in reversible light stimulation of channel opening, calcium influx and vasoconstriction.
