CRISPR / cas9 is used to insert loxP site, which is located on both sides of the region to be deleted in zebrafish. Inserting two loxP sites can permanently remove the sequence of interest through the expression of Cre recombinase, and tissue or stage specific promoter expression can be used to accurately control gene function.
The line can then be hybridized with a cre line expressing Cre recombinase under a specific promoter, or injected with a plasmid containing Cre recombinase. Cre recombinase promotes the recombination of two loxP sites, and the region between sites is removed from the genome.
One of the more difficult aspects of biology is how to study the loss of function of basic genes. Now, conditional mutagenesis by inserting loxP into zebrafish is possible. We used sequential integration of loxP sequences to create a "floxed" gene for a large number of sequence deletions in the target locus.
