Bacterial opsin is a retinal binding protein that binds photosensitive domains and ion channels or pumps. It provides light dependent ion transport, membrane potential change and sensory function for bacteria.
The strain of hemizygous mice is dynamic, fertile, normal in size, and does not show any obvious physical or behavioral abnormalities. The fragment formed by the fusion of channelrhodopsin-2 (ChR2) derived from Chlamydomonas reinhardtii and yellow fluorescent protein (YFP) was inserted downstream of the mouse thymus cell antigen 1 (Thy1) promoter of transgenic mice.
These mice can be used for optogenetic studies, rapid control of motor behavior by increasing or eliminating light, in vitro and in vivo studies of neural circuits/connections after illumination, and fluorescent labeling of Thy1 expressing cells. ChR2 acts as a blue light driven cation channel, depolarizing cells and generating action potentials. Irradiation of ChR2 expressing neurons with blue light (~ 470 nm) resulted in rapid and reversible stimulation of action potential discharge/neural activity in these cells.
